Bioavailability of Polymeric Matrix Pellet Containing Cephalexin





Abstract
The old cephalosporins such as cephalexin have short half-lives and are absorbed by carrier-mediated process usually. In order to sustain drug-release, matrices were prepared by ethylene-vinylacetate copolymer with Carbopol-940. Matrix was cutted and sieved through 840-1180§­ to evaluate in vivo bioavailability in rats. Some addition of Carbopol to matrix increased the release rate of cephalexin, On the other hand, increase in the ratio of polymer to drug decreased the release rate in pH 6.8 buffer solution. The AUC_0¡­24 atter oral administration of marix was decreased 10¡­23% compared with that of cephalexin powder. The absorption rate effectiveness(ARE) and the controlled release effectiveness(CRE) were wmployed to evaluate the pharmacokinetic characteristics of different matrices.
